I am trying to myristoylate a protein (via the N-terminal glycine) using purified recombinant N-myristoyltransferase and myristoyl-CoA. However I am having problems with solubility. I can solubilize the unmyristoylated protein in 250 mM NaCl and 25 mM HEPES at pH 8.0.
I wonder if anyone on this listserv has had a protein precipitate during myristoylation. Were you ever able to solubilize it? If so, what conditions did you use (both for myristoylation and for solubilization)?
